In this study, we developed a gold nanostar (GNS) probe for multi-modality theranostics including surface-enhanced Raman scattering (SERS) detection, x-ray computed tomography (CT), two-photon luminescence (TPL) imaging, and photothermal therapy (PTT). We performed radiolabeling, as well as CT and optical imaging, to investigate the GNS probe's biodistribution and intratumoral uptake at both macroscopic and microscopic scales. We also characterized the performance of the GNS nanoprobe for in vitro photothermal heating and in vivo photothermal ablation of primary sarcomas in mice. The results showed that 30-nm GNS have higher tumor uptake, as well as deeper penetration into tumor interstitial space compared to 60-nm GNS. In addition, we found that a higher injection dose of GNS can increase the percentage of tumor uptake. We also demonstrated the GNS probe's superior photothermal conversion efficiency with a highly concentrated heating effect due to a tip-enhanced plasmonic effect. In vivo photothermal therapy with a near-infrared (NIR) laser under the maximum permissible exposure (MPE) led to ablation of aggressive tumors containing GNS, but had no effect in the absence of GNS. This multifunctional GNS probe has the potential to be used for in vivo biosensing, preoperative CT imaging, intraoperative detection with optical methods (SERS and TPL), as well as image-guided photothermal therapy.Item Open Access Architectures for Memristor-based Storage Structures(2011) Liu, YangRapid data growth nowadays makes it more critical to reduce search time to improve the performance of search-intensive applications. However, huge data size makes it more difficult to efficiently perform search operations. Representative conventional approaches to reduce search time, such as CAM and in-memory databases, are no longer efficient because of the data explosion: CMOS-based CAM has low capacity which cannot be increased through CMOS scaling, and in-memory databases have performance degradation as data size increases. As a result, we have to exploit emerging nanotechnologies to accelerate search.
Among emerging nanotechnologies, memristors have become promising candidates to build storage structures because of high capacity, short switching time and low power consumption. However, the benefit we can obtain from these storage structures is limited by low endurance of memristors. In order to utilize the computation ability of memristors and deal with the endurance problem, we explore the design space of memristor-based storage structures.
We first propose MemCAM/MemTCAM, a configurable memristor-based CAM/TCAM design, in which we use memristors as both memory latches and logic gates. Computation ability of memristors makes it possible to perform range search and high density of memristors provides an opportunity to build MemCAM/MemTCAM with large capacity and small area. We use SPICE to model the memristor and analyze power and performance at different temperatures. The results show that it is feasible to build MemCAM and MemTCAM which have high capacity and can reduce total search time and energy consumption for search-intensive applications with huge data size.
We then propose four hybrid memristor-based storage structures, Hash-CAM, T-tree-CAM, TB+-tree, and TB+-tree-CAM, to solve the endurance problem. We use an analytical model to evaluate and compare the performance and lifetime of two software-implemented memory-based T-trees and these four hybrid storage structures. The results show that hybrid storage structures can utilize range search abilities, achieve better performance than memory-based T-trees, and improve lifetime from minutes to longer than 60 years. Furthermore, TB+-tree-CAM, a hybrid memristor-based storage structure combining T-tree, B+-tree and CAM, manages to balance between performance and lifetime and can outperform other storage structures when taking both performance and lifetime into consideration.

We explored bird macroevolution using full genomes from 48 avian species representing all major extant clades. The avian genome is principally characterized by its constrained size, which predominantly arose because of lineage-specific erosion of repetitive elements, large segmental deletions, and gene loss. Avian genomes furthermore show a remarkably high degree of evolutionary stasis at the levels of nucleotide sequence, gene synteny, and chromosomal structure. Despite this pattern of conservation, we detected many non-neutral evolutionary changes in protein-coding genes and noncoding regions. Laser interstitial thermal therapy (LITT) is an effective minimally invasive treatment option for intracranial tumors. Our group produced plasmonics-active gold nanostars (GNS) designed to preferentially accumulate within intracranial tumors and amplify the ablative capacity of LITT.Experimental design
The impact of GNS on LITT coverage capacity was tested in ex vivo models using clinical LITT equipment and agarose gel-based phantoms of control and GNS-infused central "tumors." In vivo accumulation of GNS and amplification of ablation were tested in murine intracranial and extracranial tumor models followed by intravenous GNS injection, PET/CT, two-photon photoluminescence, inductively coupled plasma mass spectrometry (ICP-MS), histopathology, and laser ablation.Results
Monte Carlo simulations demonstrated the potential of GNS to accelerate and specify thermal distributions. In ex vivo cuboid tumor phantoms, the GNS-infused phantom heated 5.5× faster than the control. In a split-cylinder tumor phantom, the GNS-infused border heated 2× faster and the surrounding area was exposed to 30% lower temperatures, with margin conformation observed in a model of irregular GNS distribution. In vivo, GNS preferentially accumulated within intracranial tumors on PET/CT, two-photon photoluminescence, and ICP-MS at 24 and 72 hours and significantly expedited and increased the maximal temperature achieved in laser ablation compared with control.Conclusions
Our results provide evidence for use of GNS to improve the efficiency and potentially safety of LITT. Liu, Yang; Crews, Deidra C; Jaar, Bernard G; Rabb, Hamid; Boulware, L EbonyBACKGROUND: High quality hospital discharge communications about acute kidney injury (AKI) could facilitate continuity of care after hospital transitions and reduce patients' post-hospitalization health risks. METHODS: We characterized the presence and quality (10 elements) of written hospital discharge communications (physician discharge summaries and patient instructions) for patients hospitalized with AKI at a single institution in 2012 through medical record review. RESULTS: In 75 randomly selected hospitalized patients with AKI, fewer than half of physician discharge summaries and patient instructions documented the presence (n = 33, 44 % and n = 10, 13 %, respectively), cause (n = 32, 43 % and n = 1, 1 %, respectively), or course of AKI (n = 23, 31 %, discharge summary only) during hospitalization. Few provided recommendations for treatment and/or observation specific to AKI (n = 11, 15 and 6, 8 % respectively). In multivariable analyses, discharge communications containing information about AKI were most prevalent among patients with AKI Stage 3, followed by patients with Stage 2 and Stage 1 (adjusted percentages (AP) [95 % CI]: 84 % [39-98 %], 43 % [11-82 %], and 24 % [reference], respectively; p trend = 0.008). AKI discharge communications were also more prevalent among patients with known chronic kidney disease (CKD) versus those without (AP [95 % CI]: 92 % [51-99 %] versus 39 % [reference], respectively, p = 0.02) and among patients discharged from medical versus surgical services (AP [95 % CI]: 73 % [33-93 %] versus 23 % [reference], respectively, p = 0.01). Communications featured 4 median quality elements. Quality elements were greater in communications for patients with more severe AKI (Stage 3 (number of additional quality elements (β) [95 % CI]: 2.29 [0.87-3.72]), Stage 2 (β [95 % CI]: 0.62 [-0.65-1.90]) and Stage 1 (reference); p for trend = 0.002). CONCLUSIONS: Few hospital discharge communications in AKI patients described AKI or provided recommendations for AKI care. Indeed, the generation of neutralizing antibodies to the 2F5 and 4E10 epitopes of HIV-1 gp41 in man may be proscribed by immune tolerance because mice expressing the V(H) and V(L) regions of 2F5 have a block in B cell development that is characteristic of central tolerance. This developmental blockade implies the presence of tolerizing autoantigens that are mimicked by the membrane-proximal external region of HIV-1 gp41. We identify human kynureninase (KYNU) and splicing factor 3b subunit 3 (SF3B3) as the primary conserved, vertebrate self-antigens recognized by the 2F5 and 4E10 antibodies, respectively. 2F5 binds the H4 domain of KYNU which contains the complete 2F5 linear epitope (ELDKWA). 4E10 recognizes an epitope of SF3B3 that is strongly dependent on hydrophobic interactions. Opossums carry a rare KYNU H4 domain that abolishes 2F5 binding, but they retain the SF3B3 4E10 epitope. Immunization of opossums with HIV-1 gp140 induced extraordinary titers of serum antibody to the 2F5 ELDKWA epitope but little or nothing to the 4E10 determinant. Identification of structural motifs shared by vertebrates and HIV-1 provides direct evidence that immunological tolerance can impair humoral responses to HIV-1.Item Open Access Multifunctional Gold Nanostars for Cancer Theranostics(2016) Liu, YangThe prevalence of cancer has increasingly become a significant threat to human health and as such, there exists a strong need for developing novel methods for early detection and effective therapy. Nanotheranostics, a combination of diagnostic and therapeutic functions into a single nanoplatform, has great potential to be used for cancer management by allowing detection, real-time tracking, image-guided therapy and therapeutic response monitoring. Gold nanostars (GNS) with tip-enhanced plasmonics have become one of the most promising platforms for cancer nanotheranostics. This work is aimed at addressing the challenges of sensitive cancer detection, metastasis treatment and recurrence prevention by combining state-of-the-art nanotechnology, molecular imaging and immunotherapy. A multifunctional GNS nanoprobe is developed with capabilities ranging from non-invasive, multi-modality cancer detection using positron emission tomography (PET), magnetic resonance imaging (MRI) and X-ray computed tomography (CT), to intraoperative tumor margin delineation with surface enhanced Raman spectroscopy (SERS) and high-resolution nanoprobe tracking with two-photon photoluminescence (TPL), as well as cancer treatment with photoimmunotherapy. The GNS nanoprobe with PET scans is particularly exceptional in detecting brain malignancies as small as 0.5 mm. To the best of our knowledge, the developed GNS nanoprobe for PET imaging provides the most sensitive means of brain tumor detection reported so far. In addition, the GNS nanoprobe exhibits superior performance as photon-to-heat transducer and can be used for specific photothermal therapy (PTT). More importantly, GNS-mediated PTT combined with checkpoint inhibitor immunotherapy has been found to trigger a memorized immunoresponse to treat cancer metastasis and prevent recurrence in mouse model studies. Furthermore, a 6-month in vivo toxicity study including body weight monitoring, blood chemistry test and histopathology examination demonstrate GNS nanoparticles’ biocompatibility. Therefore, the multifunctional GNS nanoprobe exhibits superior cancer detection and treatment capabilities and has great promise for future clinical translation in cancer management.

We have developed an innovative cancer therapy named Synergistic Immuno Photothermal Nanotherapy (SYMPHONY) by combining gold nanostars (GNS)-mediated photothermal ablation with checkpoint inhibitor immunotherapy. Our previous studies have demonstrated that SYMPHONY photoimmunotherapy not only treats the primary tumor but also dramatically amplifies anticancer immune responses in synergy with checkpoint blockade immunotherapy to treat remote and unresectable cancer metastasis. The SYMPHONY treatment also induces a 'cancer vaccine' effect leading to immunologic memory and prevents cancer recurrence in murine animal models. The PROMIS measures have previously been studied and validated in cross-sectional studies of children with nephrotic syndrome. This study was designed to longitudinally validate the PROMIS measures in pediatric nephrotic syndrome. METHODS: One hundred twenty seven children with nephrotic syndrome between the ages of 8 and 17 years participated in this prospective cohort study. Patients completed a baseline assessment while their nephrotic syndrome was active, a follow-up assessment at the time of their first complete proteinuria remission or study month 3 if no remission occurred, and a final assessment at study month 12. Participants completed six PROMIS measures (Mobility, Fatigue, Pain Interference, Depressive Symptoms, Anxiety, and Peer Relationships), the PedsQL version 4.0, and two global assessment of change items. RESULTS: Disease status was classified at each assessment: nephrotic syndrome active in 100% at baseline, 33% at month 3, and 46% at month 12. The PROMIS domains of Mobility, Fatigue, Pain Interference, Depressive Symptoms, and Anxiety each showed a significant overall improvement over time (p < 0.001). When the PROMIS measures were compared to the patients' global assessment of change, the domains of Mobility, Fatigue, Pain Interference, and Anxiety consistently changed in an expected fashion. With the exception of Pain Interference, change in PROMIS domain scores did not correlate with changes in disease activity. PROMIS domain scores were moderately correlated with analogous PedsQL domain scores. CONCLUSION: This study demonstrates that the PROMIS Mobility, Fatigue, Pain Interference, and Anxiety domains are sensitive to self-reported changes in disease and overall health status over time in children with nephrotic syndrome. The lack of significant anchoring to clinically defined nephrotic syndrome disease active and remission status may highlight an opportunity to improve the measurement of HRQOL in children with nephrotic syndrome through the development of a nephrotic syndrome disease-specific HRQOL measure.Item Open Access Sustainable Duke: Procurement & Waste(2013-04-26) Liu, Yang; Shepherd, JohnThis report examines Duke University’s Procurement Department and Waste Reduction & Recycling Department to identify opportunities for environmental improvement. With regard to waste reduction and recycling, we conducted two different waste reduction and recycling efforts to increase Duke’s 39 % recycling rate, thereby lowering the amount of trash the University sends to the landfill. We conducted a series of waste audits to better understand Duke’s to-landfill stream. Our audits revealed that 47% of the material sent to the landfill could have been recycled, and that 23% could have been composted. Second, we investigated whether Duke should consider switching to single-stream recycling (versus multiple-stream). We created models project potential impacts if Duke were to transition to single-stream recycling; the models predict that transitioning could: reduce Duke’s annual tipping fee by up to$46,568 and reduce the commensurate CO2e emissions by up to 281 tons CO2e. With regard to Procurement, we evaluated the effectiveness of Duke’s current sustainable procurement policies and practices through website information review and interviews. Additionally, we investigated the widely varying sustainable procurement approaches used by thirty other institutions of higher education to evaluate their relative merits. Our work resulted in a list of recommendations to improve upon Dukes already successful model. Regarding waste management, we suggest that Duke implements single-stream recycling and post-consumer compost collection program simultaneously on campus. Regarding procurement, we recommend that Duke moves to a more centralized purchasing system and propose several suggestions to promote sustainable purchasing.Item Open Access Synergistic Immuno Photothermal Nanotherapy (SYMPHONY) for the Treatment of Unresectable and Metastatic Cancers.(Scientific reports, 2017-08-17) Liu, Yang; Maccarini, Paolo; Palmer, Gregory M; Etienne, Wiguins; Zhao, Yulin; Lee, Chen-Ting; Ma, Xiumei; Inman, Brant A; Vo-Dinh, TuanMetastatic spread is the mechanism in more than 90 percent of cancer deaths and current therapeutic options, such as systemic chemotherapy, are often ineffective. Here we provide a proof of principle for a novel two-pronged modality referred to as Synergistic Immuno Photothermal Nanotherapy (SYMPHONY) having the potential to safely eradicate both primary tumors and distant metastatic foci. Using a combination of immune-checkpoint inhibition and plasmonic gold nanostar (GNS)-mediated photothermal therapy, we were able to achieve complete eradication of primary treated tumors and distant untreated tumors in some mice implanted with the MB49 bladder cancer cells. Delayed rechallenge with MB49 cancer cells injection in mice that appeared cured by SYMPHONY did not lead to new tumor formation after 60 days observation, indicating that SYMPHONY treatment induced effective long-lasting immunity against MB49 cancer cells.Item Open Access Toyota Motor Energy Analysis and Modeling(2016-04-27) Liu, Yang; Sun, Rui; Wang, ZirenThe automobile manufacturing industry in the U.S. uses over 800 trillion British Thermal Units (Btus) of energy and spends about $3.6 billion on it each year. As a leading automobile manufacturing corporation, our client Toyota Motor Engineering & Manufacturing North America, Inc. (Toyota) seeks opportunities to improve their accuracy in predicting manufacture energy use. With a better understanding of future energy use, Toyota will be able to identify energy efficiency opportunities and improve the overall budgeting accuracy at corporate level. To accomplish this goal, this project uses five years of historical energy consumption data to test existing energy models and identify possible improvements that can be done on a process and plant level. The products of this study include revised energy models for manufacture energy use and administrative energy use for both electricity and total energy consumption. Toyota could use our models as a reliable reference to predict future energy performance.
