Item Open Access Dectin-1 Signaling in Central Nervous System Autoimmunity(2022) Deerhake, Marion ElizabethDectin-1 is a C-type lectin receptor with diverse functions in orchestrating the innate immune response. Previous studies have primarily focused on the function of Dectin-1 in the setting of fungal infection. However, emerging evidence supports an important role for Dectin-1 in the context of autoimmunity and sterile inflammation. In this thesis, I investigated the function of Dectin-1 in central nervous system (CNS) autoimmunity. Specifically, I studied the role of Dectin-1 signaling in experimental autoimmune encephalomyelitis (EAE), a mouse model of Multiple Sclerosis (MS). Genetically modified mouse lines and immunophenotyping approaches were used to study Dectin-1 function in EAE. In addition, ex vivo studies using small molecule inhibitors and next-generation sequencing were used to perform in-depth characterization of Dectin-1 signaling mechanisms. Here I report that Dectin-1 limited EAE, while its downstream signaling molecule, Card9, promoted the disease. Myeloid cells mediated the pro-resolution function of Dectin-1 in EAE with enhanced gene expression of the neuroprotective molecule, Oncostatin M (Osm), through a Card9-independent pathway, mediated by the transcription factor NFAT. Furthermore, I found that the Osm receptor (OsmR) functioned specifically in astrocytes to reduce EAE severity. Notably, Dectin-1 did not respond to heat-killed Mycobacteria, an adjuvant to induce EAE. Instead, endogenous Dectin-1 ligands, including galectin-9, in the central nervous system (CNS) were involved to limit EAE. This thesis research reveals a mechanism of beneficial myeloid cell-astrocyte crosstalk regulated by a Dectin-1 pathway and identifies potential therapeutic targets for autoimmune neuroinflammation.

Effector T (Teff) cells, such as Th1 and Th17 cells, utilize high levels of glycolytic metabolism to fuel proliferation and function. In contrast, Treg cells require oxidative metabolism to fuel suppressive function. It remains unknown how Teff/Treg-cell metabolism is altered when nutrients are limited and leptin levels are low. We therefore examined the role of malnutrition and associated hypoleptinemia on Teff versus Treg cells. We found that both malnutrition-associated hypoleptinemia and T cell-specific leptin receptor knockout suppressed Teff-cell number, function, and glucose metabolism, but did not alter Treg-cell metabolism or suppressive function. Using the autoimmune mouse model EAE, we confirmed that fasting-induced hypoleptinemia altered Teff-cell, but not Treg-cell, glucose metabolism, and function in vivo, leading to decreased disease severity. To explore potential mechanisms, we examined HIF-1α, a key regulator of Th17 differentiation and Teff-cell glucose metabolism, and found HIF-1α expression was decreased in T cell-specific leptin receptor knockout Th17 cells, and in Teff cells from fasted EAE mice, but was unchanged in Treg cells. Altogether, these data demonstrate a selective, cell-intrinsic requirement for leptin to upregulate glucose metabolism and maintain function in Teff, but not Treg cells.Item Open Access M-HIFU inhibits tumor growth, suppresses STAT3 activity and enhances tumor specific immunity in a transplant tumor model of prostate cancer.(PLoS One, 2012) Huang, Xiaoyi; Yuan, Fang; Liang, Meihua; Lo, Hui-Wen; Shinohara, Mari L; Robertson, Cary; Zhong, PeiOBJECTIVE: In this study, we explored the use of mechanical high intensity focused ultrasound (M-HIFU) as a neo-adjuvant therapy prior to surgical resection of the primary tumor. We also investigated the role of signal transducer and activator of transcription 3 (STAT3) in M-HIFU elicited anti-tumor immune response using a transplant tumor model of prostate cancer. METHODS: RM-9, a mouse prostate cancer cell line with constitutively activated STAT3, was inoculated subcutaneously in C57BL/6J mice. The tumor-bearing mice (with a maximum tumor diameter of 5∼6 mm) were treated by M-HIFU or sham exposure two days before surgical resection of the primary tumor. Following recovery, if no tumor recurrence was observed in 30 days, tumor rechallenge was performed. The growth of the rechallenged tumor, survival rate and anti-tumor immune response of the animal were evaluated. RESULTS: No tumor recurrence and distant metastasis were observed in both treatment groups employing M-HIFU + surgery and surgery alone. However, compared to surgery alone, M-HIFU combined with surgery were found to significantly inhibit the growth of rechallenged tumors, down-regulate intra-tumoral STAT3 activities, increase cytotoxic T cells in spleens and tumor draining lymph nodes (TDLNs), and improve the host survival. Furthermore, M-HIFU combined with surgery was found to significantly decrease the level of immunosuppression with concomitantly increased number and activities of dendritic cells, compared to surgery alone. CD4+ effector T cell (Teff) (Th1 and Th17) and Treg subsets are metabolically distinct, yet the specific metabolic differences that modify T cell populations are uncertain. Here, we evaluated CD4+ T cell populations in murine models and determined that inflammatory Teffs maintain high expression of glycolytic genes and rely on high glycolytic rates, while Tregs are oxidative and require mitochondrial electron transport to proliferate, differentiate, and survive. Metabolic profiling revealed that pyruvate dehydrogenase (PDH) is a key bifurcation point between T cell glycolytic and oxidative metabolism. PDH function is inhibited by PDH kinases (PDHKs). PDHK1 was expressed in Th17 cells, but not Th1 cells, and at low levels in Tregs, and inhibition or knockdown of PDHK1 selectively suppressed Th17 cells and increased Tregs. This alteration in the CD4+ T cell populations was mediated in part through ROS, as N-acetyl cysteine (NAC) treatment restored Th17 cell generation. Moreover, inhibition of PDHK1 modulated immunity and protected animals against experimental autoimmune encephalomyelitis, decreasing Th17 cells and increasing Tregs. Together, these data show that CD4+ subsets utilize and require distinct metabolic programs that can be targeted to control specific T cell populations in autoimmune and inflammatory diseases.Item Open Access Sentinel Functions of Tissue Resident Macrophages During Early Fungal Infections(2020) Xu, ShengjieInvasive fungal infections result in high morbidity and mortality. Host organs targeted by fungal pathogens vary depending on the route of infection and fungal species encountered. Across different tissues in hosts, tissue-resident macrophages (TRMs) are one of the most abundant cells of the innate immune system and are in the front-line sensing invading fungi and elicit anti-fungal response. Neutrophil recruitment to the site of infection is one of the hallmarks of immune defense. In this thesis I found that TRMs are potent producers of neutrophil chemokine CXCL2. I used CXCL2 reporter animal model and uncovered two layers of TRMs heterogeneity: differential CXCL2 expression among TRMs of various tissues and transcriptional heterogeneity within alveolar macrophages (AMs).
Different fungal infection models were utilized to examine TRMs in tissues with specific fungal tropism. Peritoneum macrophages, kupffer cells, kidney macrophages and alveolar macrophages were all capable of producing CXCL2 at the early hours post fungal infection. On the contrary, spleen macrophages and microglia were unable to produce CXCL2. Chromatin status of TRMs from naïve mice revealed that CXCL2-non-expressing TRMs also had less accessible Cxcl2 promoter than CXCL2-expressing TRMs, suggesting the regulation of Cxcl2 in TRMs at the chromatin level.
Among the CXCL2-expressing TRMs, heterogeneous transcriptional states were identified within alveolar macrophages (AMs) populations. Although Cxcl2 promoter in AMs are constantly accessible, some AMs could not produce CXCL2 protein regardless the fungal inocula, spatial distribution or ontogenic origins. In addition to be a chemokine, CXCL2 was also a biological marker that differentiated AM cell states based on unbiased single cell RNA-seq. CXCL2+ and CXCL2- AMs expressed pro- and anti-inflammatory molecules, respectively, as well as distinct metabolic profiles and phagocytic capacity. CXCL2-divided AMs cell states also have distinct chromatin status enriched with differential transcription factor motifs. Long-term study of AM populations by ex vivo or in vivo transfer revealed CXCL2+ pro-inflammatory states were plastic and could be reset in AMs. However, one exception was the anti-inflammatory C1q molecule, expressed only by the pre-CXCL2- AMs. Promoters of C1q genes become accessible during early fungal infection only in CXCL2- AMs leading to their long-term gene expression.
Fungal infections revealed quite heterogeneous CXCL2 responses both among TRMs and within AM populations. I used biologic reporter system and multiple system biology approaches to expand our knowledge of TRMs diversity and AM cell states. Particularly, my thesis demonstrated the co-existence of heterogenous AMs states with pro- and anti-inflammatory profiles, distinguished by CXCL2 during pulmonary Cryptococcus infection in vivo. The simultaneous appearance of functionally opposite AM states in the lung implies the balance of inflammation and tolerance even within a highly defined population of AMs.

Here we found that osteopontin (OPN) skewed this balance during pathogenic conditions such as infection and autoimmunity. Notably, two isoforms of OPN exerted distinct effects in shifting this balance through cell-type-specific regulation of apoptosis. Intracellular OPN (iOPN) diminished the population size of myeloid progenitor cells and myeloid cells, and secreted OPN (sOPN) increase the population size of lymphoid cells. The total effect of OPN on skewing the leukocyte population balance was observed as host sensitivity to early systemic infection with Candida albicans and T cell-mediated colitis. Our study suggests previously unknown detrimental roles for two OPN isoforms in causing the imbalance of leukocyte populations.Item Open Access The AIM2 Inflammasome Is Activated In Astrocytes During EAE(2021) Barclay, William ElliotThe inflammasomes are a group of pattern recognition receptors (PRRs) with unique characteristics and critical to innate immunity by translating microbial and damage signals into inflammation. While early investigation into inflammasomes focused on their ability to respond to invading pathogens, now inflammasomes are known to collectively respond to a broad range of sterile damage signals. Indeed, several inflammasomes, and most notably the NLRP3 inflammasome, are known to promote pathogenesis of several autoinflammatory conditions, including the autoimmune neuroinflammation which is modeled in the mouse model of multiple sclerosis (MS), experimental autoimmune encephalomyelitis (EAE). While this association of inflammasomes with EAE is well established, the timing, tissue localization, and cell-specificity of inflammasome activation in the central nervous system (CNS) remains poorly understood during disease. Thus, this dissertation details our investigation into the specific sites and timing of inflammasome activation during EAE, as well as determining which inflammasome is activated in the CNS during disease. The interrogation of inflammasome activation in vivo during disease states required our use of genetically modified mice with fluorescent-protein tagged inflammasome adaptor ASC (ASC-Citrine), which serves as a reporter of active inflammasomes. We identified in situ inflammasome activation in specific CNS cell types of these mice using antibody-based immunofluorescent staining techniques and confocal microscopy, and confirmed the result by genetically modified mice, which allowed cell-specific expression of ASC-Citrine. Further, we used mice genetically deficient in inflammasome components ASC and AIM2 to investigate the involvement of these proteins in disease development. Our study concluded with several insights. Firstly, inflammasome activation occurs in the antigen draining lymph nodes prior to symptom onset during EAE, but inflammasome activation occurs more significantly in the spinal cord at 30 days post induction (dpi) of EAE. Spinal cord inflammasome activation during EAE occurs selectively in radioresistent cells, mainly in astrocytes. Further, this astrocyte inflammasome is dependent on AIM2, and does not result in outcomes, which are canonically observed in myeloid cells, such as release of the inflammatory cytokine, IL-1β. Indeed, AIM2 limits EAE development. Lastly, astrocyte inflammasomes differ morphologically from the traditional structure (known as the “ASC speck”) as seen in macrophages. This dissertation thus expands our understanding of inflammasome activation during EAE, and introduces new areas of investigation into the AIM2 inflammasome during EAE and inflammasome activation in astrocytes.
Item Unknown The Role of IFN-γ and STAT1 Signaling in Neuronal Excitability and Behavior(2023) Clark, Danielle NicoleThe IFN-γ/STAT1 response is an immune signaling pathway well known for its potent pro-inflammatory and anti-viral functions. However, IFN-γ/STAT1 signaling also impacts many homeostatic and pathological aspects in the central nervous system, beyond its canonical role in controlling intracellular pathogens. IFN-γ can modulate neuronal excitability, synaptic pruning, and gene expression of pathways associated with neurodevelopmental disorders, including autism spectrum disorder (ASD) and schizophrenia (SZ). Surprisingly, the IFN response was recently identified as the most highly enriched pathway in brains of individuals with ASD and SZ. Children born to mothers who are hospitalized for infection during pregnancy are at a higher risk of developing ASD, and mouse models demonstrate that elevating cytokines during embryonic neurodevelopment cause ASD-like phenotypes. While microglia are thought to be the major targets of IFNs in the brain, neurons can respond to IFNs and require physiological levels of IFN-γ for proper function. The IFN-γ/STAT1 pathway is rapidly activated then deactivated to prevent excessive inflammation; however, neurons utilize unique IFN-γ/STAT1 activation patterns, which may contribute to the non-canonical neuron-specific downstream effects. We hypothesized that pathological IFN-γ signaling in neurons leads to neuronal dysfunction and behavioral deficits through non-canonical STAT1 signaling. Using primary neuron cultures, we demonstrated that developing neurons have differential STAT1 activation downstream of physiological versus pathological IFN-γ. Physiological levels of IFN-γ caused brief and transient STAT1 activation, while high pathological levels of IFN-γ caused robust and prolonged activation of STAT1 in neurons, but not in microglia or astrocytes. To determine the effects of prolonged STAT1 activation in vivo, we developed a novel mouse model in which STAT1 signaling is prolonged in neurons. These mice displayed hyperactive behavior and neural hypoactivity, which are common comorbidities of neurodevelopmental disorders like ASD and attention deficit hyperactivity disorder (ADHD). Moreover, we demonstrated that this phenotype is neuron specific, as mice with prolonged STAT1 activation in microglia did not have behavior deficits. Our findings suggest pathological activation of the IFN-γ/STAT1 pathway contributes to neuronal dysfunction through non-canonical STAT1 activation. Overall, the IFN-γ/STAT1 pathway is critical for normal neurodevelopment and neuronal function in adulthood and provides new insight into a neuron specific neuroimmune mechanism which may contribute to the pathophysiology of neurodevelopmental disorders.

Host organs targeted by fungal pathogens vary depending on the route of infection and fungal species encountered. Cryptococcus neoformans infects the respiratory tract and disseminates throughout the central nervous system. Candida albicans infects mucosal tissues and the skin, and systemic Candida infection in rodents has a tropism to the kidney. Aspergillus fumigatus reaches distal areas of the lung once inhaled by the host. Across different tissues in naïve hosts, tissue-resident macrophages (TRMs) are one of the most populous cells of the innate immune system. Although they function to maintain homeostasis in a tissue-specific manner during steady state, TRMs may function as the first line of defense against invading pathogens and may regulate host immune responses. Thus, in any organs, TRMs are uniquely positioned and specifically programmed to function. This article reviews the current understanding of the roles of TRMs during major fungal infections.Item Unknown Zfp335-Mediated Regulation of T cell Development(2022) Ratiu, JeremyT cells are a critical arm of the adaptive immune system which function to coordinate and orchestrate complex immune reactions, as well as, kill damaged and infected cells. Production of a diverse peripheral T cell compartment requires massive expansion of the bone marrow progenitors that seed the thymus. There are two main phases of expansion during T cell development, following T lineage commitment at the DN2 stage and following successful rearrangement and selection for functional TCRβ chains in DN3 thymocytes, which promotes development of DN4 cells to the DP stage. Signals driving expansion of DN2 thymocytes are well studied, however, factors regulating the proliferation and survival of DN4 cells remain poorly understood.
E proteins are transcription factors which have been shown to play essential non-redundant roles throughout T cell development. The functions of E proteins in T cell development include, enforcing T lineage commitment, promoting proper TCR rearrangements, regulating developmental progression and functional checkpoints, and coordinating complex transcriptional networks underpinning developmental progression. Due to the large number of genome-wide binding sites and massive number of genes regulated by E proteins, their numerous functions are poorly understood. The goal of this dissertation is to determine the role of the E protein-regulated transcription factor Zfp335 in T cell development.
We utilized conditional deletion models to determine the role of Zfp335 in early and late stages of conventional and unconventional T cell development. Through these efforts we uncovered we uncover an unexpected link between the transcription factor Zfp335 and control of the cGAS/STING pathway for sensing cytosolic DNA in post-β-selection DN4 thymocytes. The absence of Zfp335 drives cGAS/STING-dependent death of DN4 cells. Zfp335 controls survival by sustaining expression of Ankle2, which in turn regulates the activity of Baf to suppress cGAS/STING-dependent cell death. Additionally, genetic ablation of Zfp335 precludes the development of unconventional iNKT cells due to STING-independent cell death following lineage commitment along with preventing effector differentiation of surviving cells.
Our studies also uncovered an additional cGAS/STING-independent role in the terminal maturation of conventional αβ T cells. The absence of Zfp335 prevents the establishment of a naïve T cell compartment, inhibits differentiation of CD4 T cells and promotes the developmental acquisition of an effector program in CD8 T cells. Using in vivo and ex vivo genetic manipulation combined with detailed bioinformatic analyses we show that Zfp335 functions to promote T cell development, maturation, and effector differentiation through the regulation of a small but essential set of genes.
To our knowledge, these studies detail the first described role for cGAS/STING in T cell development and strongly suggest a transcriptional mechanism downstream of Zfp335 which coordinates genome-wide alterations to chromatin compaction required for proper establishment of conventional and unconventional T cell pools. Together, these studies will provide a novel framework for understanding the life and death of developing T cells and may uncover novel pathways for enhancing the efficacy of T cell-based therapeutics.
