Item Open Access Characterizing sleep-wake cycles in dogs (Canis lupus familiaris) using daytime activity levels, sex, and temperament: a longitudinal comparison(2024-04-12) Sandberg, EmilyAcross many species, sleep patterns are associated with variables such as age, sex, daytime activity levels, and temperament. Yet, current research lacks an in-depth characterization of dog sleep patterns and how they vary according to these variables during the critical developmental period of young puppyhood. Such studies are crucial in order to establish dogs as a model organism for studies of sleep and for additional applications in the realms of dog welfare and training. In the present study, we characterized how often and for how long young dogs wake, as well as their barking patterns during sleep. We evaluated sleep behaviors for dogs aged 8-18 weeks and determined longitudinal patterns using a sample of Canine Companions service-dogs-in-training (N=21). Video recordings of dogs were analyzed using a novel coding scheme to determine duration and frequency of awake bouts and barking. Mixed-effects logistic regression models reveal that awake-bout length (minutes) and frequency did not vary significantly by weeks of age, daytime activity levels, or temperament. However, we did find significant sex differences in awake bout length (p<0.009). These results suggest distinct daytime and nighttime temperaments, as well as the importance of this developmental period for developing adultlike sleep patterns. Further study is required to examine sleep behaviors in puppies beyond 18-weeks to better understand how adultlike patterns emerge and the stability of the patterns observed in this study.Item Open Access The dynamics of successive induction in larval zebrafish.(J Exp Anal Behav, 2010-09) Staddon, JER; MacPhail, RC; Padilla, SCharles Sherrington identified the properties of the synapse by purely behavioral means-the study of reflexes-more than 100 years ago. They were subsequently confirmed neurophysiologically. Studying reflex interaction, he also showed that activating one reflex often facilitates another, antagonistic one: successive induction, which has since been demonstrated in a wide range of species, from aphids to locusts to dogs and humans. We show a particularly orderly example in zebrafish (Danio rerio) larvae; the behavior (locomotion) of larvae is low in dark and intermediate in light, but low in light and substantially higher in dark when dark followed light. A quantitative model of a simple dynamic process is described that readily captures the behavior pattern and the effects of a number of manipulations of lighting conditions.Item Open Access The Growth and Activity of Genetically Diverse Prochlorococcus(2013) Lin, YajuanWhile much is known about the abundance and genetic diversity of environmental microbial communities, little is known about their taxon-specific activity. In this thesis I address this gap using a model marine microbe, the cyanobacterium Prochlorococcus spp., which numerically dominates tropical and subtropical open oceans and encompasses a group of genetically defined clades that are ecologically distinct. Ribosomal RNA is a promising indicator of in situ activity because of its essential role in protein synthesis as well as its phylogenetic information, which could be used to distinguish clades among mixed populations. Here I show that, in a laboratory system the specific growth rate of representative Prochlorococcus strains could be quantitative predicted from cellular rRNA content (assessed by RT-qPCR), cell size (assessed by flow cytometry) and temperature. Applying this approach in the field, I show unique clade-specific activity patterns for Prochlorococcus. For example, vertically within the euphotic zone, eHL-II activity is strongly impacted by light and is consistent with patterns of photosynthesis and on a horizontal transect from Hawaii to San Diego, eHL-I and eHL-II activities exhibit significant transitions and appear to be regulated by temperature, nutrient and vertical mixing gradients. Using ribosomal tag pyrosequencing of DNA and RNA, I have extended our observation to the Eubacterial community and described the biomass distribution (rDNA) and activity (rRNA) patterns from two representative depths (25 and 100 m) at a well-studied oligotrophic ocean station. These results show that for some populations the abundances and activities are significantly uncoupled, which suggests substantial top-down controls or physical transport processes. Further exploring the taxon-specific activity patterns along with abundances and environmental variables across time and space is essential to better understanding the dynamics of a complex microbial system as well as predicting the consequences of environmental change.
Item Open Access Walking All over COVID-19: The Rapid Development of STRIDE in Your Room, an Innovative Approach to Enhance a Hospital-Based Walking Program during the Pandemic.(Geriatrics (Basel, Switzerland), 2021-11) Hughes, Jaime M; Bartle, John T; Choate, Ashley L; Mahanna, Elizabeth P; Meyer, Cassie L; Tucker, Matthew C; Wang, Virginia; Allen, Kelli D; Van Houtven, Courtney H; Hastings, Susan NicoleHospitalization is common among older adults. Prolonged time in bed during hospitalization can lead to deconditioning and functional impairments. Our team is currently working with Department of Veterans Affairs (VA) medical centers across the United States to implement STRIDE (assiSTed eaRly mobIlity for hospitalizeD older vEterans), a hospital-based walking program designed to mitigate the risks of immobility during hospitalization. However, the COVID-19 pandemic made in-person, or face-to-face, walking challenging due to social distancing recommendations and infection control concerns. In response, our team applied principles of implementation science, including stakeholder engagement, prototype development and refinement, and rapid dissemination and feedback, to create STRIDE in Your Room (SiYR). Consisting of self-guided exercises, light exercise equipment (e.g., TheraBands, stress ball, foam blocks, pedometer), the SiYR program provided safe alternative activities when face-to-face walking was not available during the pandemic. We describe the methods used in developing the SiYR program; present feedback from participating sites; and share initial implementation experiences, lessons learned, and future directions.
